Is it possible to be able to get full kill when using the engine dj software on the mixstream pro as a standalone. When having all 3 EQ knobs fully down, sound still comes out. Being able to get full gives you much better freedom when mixing with Isolating mode.

No, EQ type is not part of the Mixer Preferences in the Mixstream Pro. It might be achievable by a future update if the feature request gets enough votes. it might also be a decision to keep a difference between the lower and the higher price range devices.
The Prime 4, Prime 2 and Prime Go do support this, so if you really want it you could consider the upgrade.
